Previously we described for the intestine, a circadian rhythm in the labelled nuclei/crypt, mitotic figures/crypt and disintegrations per minute/crypt (Exp. Cell Res. 58, 159, 1969; Experientia 26, 1321, 1970). This rhythm was completely reversed by reversing the light cycles (J. Cell Physiol., 78, 121, 1971). The present investigation tested the cell cycle times of the proliferative crypt cells by the percent labelled mitosis method. The results of the percent labelled mitosis when proliferation was at its peak (03.00 hours), showed the cell cycle times to be 11.1 hours. The generation time obtained when the curve was started at the nadir of proliferation was 13.1 hours. The difference was primarily due to a shortening of G1 phase of the cell cycle. The number of proliferative cells/crypt was calculated at the peak and nadir and found to be 114 and 82, respectively. Possible mechanisms are discussed.
