Peripheral blood monocyte-derived macrophages from genetically resistant and susceptible cattle to intracellular parasites were evaluated for their ability to control the intracellular growth of virulent M. bovis. Bactericidal assays using a 10:1 multiplicity of infection were performed to study bacterial phagocytosis and bacterial multiplication at 0 and 24 h post-inoculation. The rate of bacterial phagocytosis was 1.4 ± 0.17 and 2.1 ± 0.54 bacteria/macrophage, whereas bacterial multiplication was 165% and 263.7% in macrophages from resistant and susceptible cattle respectively. These results confirm that macrophages from resistant and susceptible cattle differ significantly (P<0.01) in controlling intracellular multiplication of virulent M. bovis. |
