The oral bioavailability of BCS (biopharmaceutics classificationsystem) class II drugs with poor solubility andreasonable permeability is limited by the drug dissolutionstep from drug products. Though prodrug approach is anexciting way of improving the oral bioavailability, it requiresextensive studies to establish the safety profile ofprodrugs in humans. In view of the increasing market shareof oral drug products, a variety of technologies are developedto enhance the oral bioavailability of poorly solubledrugs using the excipients with approved or GRAS (generallyregarded as safe) status. The present review describesthe main technologies such as micronization, nanosizing,crystal engineering, solid dispersions, cyclodextrins, solidlipid nanoparticles and other colloidal drug delivery systemswith a few relevant research reports. |
