Contents: A qualitative test for p-coumarate hydroxylase, measuring solution reductant levels one hour after addition of p-coumaric acid to a CaCO3 buffered nutrient solution containing Fe-stressed sorghum, was examined as a possible biochemical assay for sorghum cultivar Fe-efficiency screening. P-coumarate hydroxylase is an important enzyme in the biochemical synthesis of caffeic acid, while caffeic acid has been identified as a major reductant released by certain Fe-efficient plants while under Fe-stress. All ten sorghum cultivars tested in this study contained the enzyme and could not be differentiated for Fe-efficiency on this basis. The enzyme was highly active during testing on plants grown in CaCO3 buffered nutrient solution (pH 7.5), but the enzyme appeared to be inactivated in pH 3 nutrient solution.
