Purity of high performance liquid chromatographic solvents was investigated under buffered conditions, Phosphate buffer was used in this analysis because it suits a wide pH range, The buffered mobile phase had a higher absorbance than either acetonitrile or water solvent, therefore, the liquid chromatographic separation had a tremendous negative drift when the composition of solvent changes in a gradient system from 100% buffer to 100% acetonitrile, the less absorbed acetonitrile solvent gives the larger negative drift, This makes the analysis extremely difficult to control the baseline and depends on the absorbance of the organic solvent in addition to the difference in the mobile phase absorbance, the trace impurities in acetonitrile solvent serve as interferences, Different lots of acetonitrile from different venders were used to provide different levels of impurity interferences in this investigation.
