An improved isolation procedure for a cytolytic material ("aponin") from the blue green alga, Gomphosphaeria aponina is described. The cytolytic activity of aponin toward G. breve, an unarmored dinofiagellate associated with Florida red tides, was assayed. Cytolytic activity of the crude aponin showed no change following incubation for one hour at various temperatures over the range 30-110°C. Aponin was acid stable, and base labile, on the basis of incubation at 30°C in distilled water at definite pH for 48 hours prior to bioassay with G. breve. In sea water, the cytolytic activity remained constant for 1-10 days, following which the cytolytic activity decreased. Preliminary purification was effected using silica gel chromatography, followed by permeation gel chromatography (Sephadex LH-20).
