Contents: The initial adsorption behavior of fibrinogen on artificial surfaces was investigated with respect to adhesion of platelets and the in vivo anthhrombogenicity of materials. The saturated amount and the initial rate of fibrinogen adsorption were not very high on poly(γ-benzyl L-glutamate) (PBLG), a thrombogenic material. However, the PBLG surface, in the initial adsorption stage with fibrinogen, was detected to bind with a relatively high amount of anti-fibrinogen antibodies, which indicates a lack of conformational changes of fibrinogen adsorbed on the PBLG surface, although the number of platelets adhering to PGLG was not very high in in vitro experiments. Fibrinogen molecules adsorbed on PBLG may cause effective platelet aggregation, or coagulation factors may participate in thrombus formation on the PBLG surface. On the other hand, relatively large number of platelets adhered to polyether urethane nylon, which resulted in good in vivo antithrombogenicity and showed no remarkable adsorption behavior of fibrinogen. Thus, the number of platelets adhering in vitro to these surfaces did not correspond directly to in vivo antithrombogenicity.
