Contents: Daily rhythmic changes in the concentration and 3H-lysine incorporation of histones and nonhistones proteins of rat liver nuclei The total amount of nuclear proteins in rat liver changes rhythmically during 24 hrs: A maximum is reached at 24.00 hr, a minimum at 16.00 hr. 3H-Lysine incorporation into nuclear proteins is highest between 12.00-16.00 hr and lower between 20.00-24.00 hr. The histone-DNA ratios show only small differences between 1.065 at 16.00 hr and 1.196 at 20.00 hr. 3H-Lysine incorporation into total histones is maximal at 16.00-20.00 hr and minimal at 20.00-24.00 hr. The F1- (lysine-rich) fraction of the histones and the other histones both change in their relative concentrations: The maximum of the lysine-rich fraction occurs between 20.00 and 24.00 hr whereas the rest of the histones accumulates 4 hrs earlier. 3H-Lysine incorporation into the F1-fraction exhibits a pronounced daily rhythmicity with a maximum at 20.00 hr and a minimum at 24.00 hr. The incorporation into the other histones is less and the range of the rhythmicity lower compared to the lysine-rich fraction. The amount of nonhistone proteins also changes rhythmically during 24 hrs: A maximum protein-DNA ratio of 0.699 is observed at 24.00 hr, a minimum ratio of 0.523 at 20.00 hr. 3H-Lysine incorporation into the nonhistone fraction is maximal between 12.00 and 20.00 hr and minimal at about 2.00 hr. The RNA content in the chromatin shows a maximum at 8.00 hr.
