With the advent of new pharmaceutical agents designed to be delivered in aerosol form to the respiratory tract for local or systemic benefit, the issue of acceptable dose distribution related to generation, delivery, and breathing becomes an important consideration in achieving maximal benefit. This paper examines the definition of aerosol dose for pharmaceutic agents, considers the necessary specification of dose distribution, and reviews data of pharmaceutic aerosol delivery to estimate the factors responsible for variability. It is concluded that the required degree of dose distribution specification is agent dependent, and that efforts to design aerosol delivery systems for pharmaceutical agents must have the desired dose specification established at the beginning of the design effort. |
