This paper proposes the use of the randomization distribution in the calculation of adjusted p-values, point estimates and confidence intervals following a sequential clinical trial. The adjusted p-value is calculated by repeating the sequential analysis process for each (or a random sample) of the possible random allocations of patients to treatments, and comparing the obtained results with the actual result on the basis of sample size at termination. Adjustments to the point estimate and confidence interval follow from the adjusted p-value. This procedure is conceptually simple and can be applied to all sequential trials regardless of the type of sequential boundary or the distribution of the patient populations. |
