The interference of various concentrations of soluble protein and organic acids with chloride (Cl-) determinations using a specific ion electrode was evaluated. The organic acids, malic and citric, provided 4 to 5 times the Cl- interference as did soluble protein at equivalent concentrations. A definite interaction of the two interfering substances existed which also led to erroneous estimations of the actual Cl- concentration. A comparison of corn leaves dry ashed in a basic media versus water extracted resulted in 4 to 5 times hi gher Cl-values in the water extract. The results of this experiment suggest that water extraction of plant tissue for Cl- determinations will result in an overestimation of the actual Cl-value when using the specific ion electrode. Dry ashing the sample in a basic media, then extracting with hot water results in reliable estimates of the Cl- concentration. |
